Positional cloning

The positional cloning includes methods for identification of unknown DNA sequences in the immediate vicinity of a known DNA sequence. In contrast to functional cloning , positional cloning is based on the search for an adjacent DNA sequence on the same DNA molecule. This adjacent DNA sequence may belong to the same operon as the known sequence. The DNA is purified by DNA isolation and determined by primer walking (a method of DNA sequencing ).
