UDS test

The UDS test is a test procedure with which genotoxic damage can be detected indirectly through induced repair mechanisms. The acronym UDS stands for " u nscheduled D NA s ynthesis" or " u nplanmäßige (unscheduled) D NA S ynthesis". However, it must be taken into account that not all genotoxic substances also induce DNA repair (e.g. certain heavy metals), so that false-negative results can possibly occur.

For the UDS test, the test substance and [ 3 H] -thymidine ( tritium - thymidine ) are added to hepatocytes . If the DNA is damaged by the test substance and repair mechanisms become active, [ 3 H] thymidine is incorporated.

To evaluate the UDS test, the cells are fixed and subjected to autoradiography . The more intense the blackening of a photographic film, the greater the DNA damage.

For example, UDS could be detected in analyzes with various antibiotics from the fluoroquinolones group such as ciprofloxacin .
