LA 378 runs through Moss Bluff, an unincorporated area north of Lake Charles, and Westlake, a small incorporated area northwest of Lake Charles.

It starts at Hwy 14 (also known as Hwy 171) and runs through the center of Moss Bluff. The Moss Bluff stretch is named Sam Houston Jones Parkway. The highway follows Davis Road into Westlake, then proceeds down Phillis Rd., Westwood Rd., and Sampson St. It ends at I-10.
