On August 1, 1737, Klose filled the first physics position in the city of Heilbronn, which had previously been vacant two years, and was sworn in a few days later. In 1738 he received citizenship and was the owner of the Trappensee property . Especially from the first years as a city doctor, several complaints against Klose because of neglect of his duties have come down to us. Strangers threw him window panes several times. From 1751 onwards he is mentioned with the title of court counselor. In 1755 he switched to the Margrave of Baden-Durlach as a personal physician.
