Using the GAL4 / UAS system (also UAS-GAL4 system ) can any cloned gene selectively in specific cells or tissues expressing be. GAL4 is a yeast-specific transcription factor that is under the control of a weak promoter and codes for the GAL4 protein. Another promoter or enhancer is required for expression . The GAL4 protein binds specifically to the so-called UAS (Upstream Activating Sequence), which activates a target gene located downstream. This can e.g. B. the gene for the green fluorescent protein GFP , or a gene to be examined.

The GAL4 / UAS system was developed by Andrea Brand and Norbert Perrimon at Harvard Medical School for the model organism Drosophila melanogaster and first published in Development in 1993 . It is often used in conjunction with molecular biology methods such as the Enhancer Trap method.
