PDE-4 inhibitors

from Wikipedia, the free encyclopedia

Phosphodiesterase-4 inhibitors (also phosphodiesterase-IV-inhibitors , PDE-4-inhibitors ) are substances that inhibit the enzyme phosphodiesterase-IV . Phosphodiesterase IV breaks down the second messenger cAMP . PDE-4 inhibitors therefore increase the concentration of intracellular cAMP. The enzyme is found in the lungs, among other things.

The prototype of the phosphodiesterase 4 inhibitor is rolipram . PDE-4 inhibitors have anti-inflammatory effects and have been a. investigated in the areas of application of COPD , bronchial asthma , depression and multiple sclerosis . So far, two active ingredients have been approved as medicinal products: roflumilast (Daxas ® ) and apremilast (Otezla ® ). The development of cipamfylline has been discontinued.
