Cyanophilia
In fungi, cyanophilia is the blue coloration of plasma parts or cell walls of basidia , cystidia , hyphae or spores due to cotton blue . If the structures are clearly colored blue, they are called cyanophilic (loving the blue dye), if the structures are not colored blue, they are called acyanophilic .
Spore staining
The cyanophilicity check is particularly often used for fungal spores. Spores are cyanophilic when they turn the outer wall or the spore ornament blue to blue-violet with cotton blue. The color solution consists of 0.17 g of cotton blue in 100 ml of concentrated lactic acid. Only fresh material should be used for the test. The Clemençon staining is done as follows:
A fragment from the hymenium is placed directly in the color solution on a slide. (A spore-shedding preparation is unsuitable, as immature spores are also required for the assessment.) The preparation is carefully heated until it boils and then examined under the microscope. Assessment can sometimes be difficult as the contents of the spores can also stain. Skillful focusing can be used to determine whether the spore wall is really stained.
Hyphae staining
A trama or hymenium section is placed on a slide and a cotton blue lactophenol solution is then dripped onto it. Then it is heated to a boil. The trama- or hymenium "tissue" should turn blue if the reaction is positive. The cotton blue / lactophenol color solution consists of: 20 g phenol, 20 g lactic acid, 40 g glycerine, 0.05 g cotton blue and 20 ml distilled water .
