The State Route begins on Delaware State Route 41 in Hockessin and ends after eleven kilometers in Wilmington on a business route on US Highway 13 .

After branching off from DE 41, Delaware State Route 48 runs east and meets Delaware State Routes 100 and 141 in the western suburbs of Wilmington .
