He was a son of Wolf von Gemmingen († 1555) and Anna Marschalk von Ostheim († 1569). His teachers were the pastors Franciscus Irenicus and Wolfgang Busius in Gemmingen . Later he came to the court of Pfalz-Zweibrücken and became director of the knight canton of Kraichgau.

Together with his brother Pleikard (1536–1594), who was still underage when his father died , he inherited the father's property. The brothers later divided up the inheritance, which gave Dietrich Gemmingen a hearing. In 1573 he also acquired Gut and Schloss Filseck and Weilerberg in the Oberamt Göppingen from Balthasar Moser von Filseck and Weilerberg , which his heirs sold again in 1596.

He was buried in the old church in Gemmingen, his grave slab is preserved in the garden of the Gemmingen lower castle.
