The FLAG tag is a protein tag that is used in protein purification and protein characterization of recombinant proteins .

The FLAG tag has the amino acid sequence D Y K DDDDK. This increases the mass by 1012 Da . It can be attached to the N -terminus or the C -terminus of the recombinant protein and also as a TAP tag , but some antibodies only recognize and bind it to the N -terminus, e.g. B. M1 / ​​4E11. Occasionally the tyrosine in the FLAG tag is sulphated , which can impair antibody binding. As with other protein tags, a proteaseInterface between the recombinant protein and the FLAG tag can be inserted in order to proteolytically cleave off the FLAG tag after purification.
