He was able to expand his knowledge on a long trip to Italy. Guillemain became a student of the well-known violinists Giovanni Battista Somis and Jean-Marie Leclair .

From 1729 he was a member of the orchestra at the Lyon Opera, and in 1734 he became concertmaster of the Royal Academy in Dijon. In the following years he traveled to Paris, where he was active in many areas of music, both as a composer and as a violinist. In 1759 he entered the royal service as a violinist. In the course of his life he gained important positions at the court of Versailles. Obviously cornered by overwhelming debts, he took his own life with 14 knife wounds on the drive from Paris to Versailles near Chaville .

The contemporary Louis-Claude Daquin wrote of famous people in his letter: “When one speaks of a man full of genius, vivacity and fire, one cannot avoid mentioning the royal violinist Mr. Guillemain. His hand is bubbly and no difficulty can be held back. This fantastic artist is one of the most fruitful of the great masters, his works are piquant and full of beauty. "
