Indirect Immunoperoxidase Assay

The indirect immunoperoxidase assay (English: 'indirect immunoperoxidase test', IPA) is a virological method for the detection and quantification of viruses . The IPA is a virological variant of immunohistochemistry in a cell-based immunoassay and was used in virological diagnostics .

principle

The IPA is mainly used for viruses without a cytopathic effect ( non-lytic viruses) to determine a titer . In contrast to lytic viruses, non-lytic viruses cannot be measured in a plaque assay , which is why an IPA or other TCID 50 assays are usually used for this, e.g. B. with the detection of the human coronaviruses SARS -CoV, CoV 229E and CoV OC43, or with the hepatitis A virus . In IPA, cell cultures are inoculated with dilution series of the viruses, after a period of viral replication the cells are fixed , provided with antibodies and detected by immunostaining and counted under a microscope. Instead of an unknown virus concentration, the IPA can also - if the virus concentration is known - be used to determine antibodies, e.g. B. in the human cytomegalovirus or rotaviruses .
