The melanocortin-4 receptor (short: MC4R ) belongs to the group of melanocortin receptors and is a G-protein-coupled receptor . It is expressed in the brain (especially in regions of the hypothalamus near the organum vasculosum laminae terminalis ) and in the intestinal tissue, but not in the placenta , the adrenal cortex or in melanocytes. It is activated by α-MSH , β-MSH , γ-MSH and by ACTH and is involved in vegetative regulation of energy homeostasis. For example, it raises the normal body temperature, but suppresses the fever reaction . It also suppresses the feeling of hunger (e.g. in the context of tumor cachexia) and influences the energy consumption in the metabolism and increases sexual appetite.

The protein for MC4R is encoded in an exon on chromosome segment 18q22 and contains 333 amino acids .

Mutations

Mutations in the MC4R can also occur in heterozygotes with an autosomal predisposition to obesity .
