The melanocortin receptor 1 ( MC1R ) is one of the melanocortin receptors . In humans, it is encoded on chromosome 16 gene locus q24.3 and is a transmembrane G protein-coupled receptor . MC1R is expressed in the melanocytes and controls both hair color and skin tan. There are over 30 different receptor alleles that determine skin type (sensitivity to skin radiation and sunburn). A special mutation leads to hereditary susceptibility to malignant melanoma .

The locus of this receptor is called the extension locus .
