NaSSA

NaSSA is the abbreviation for English noradrenergic and Specific Serotonergic Antidepressant ( dt .: noradrenergic and specific serotonergic antidepressant), a group of antidepressant psychotropic drugs . They are among the so-called "newer antidepressants".

These tetracyclic active ingredients bind to binding sites ( receptors ) for the monoamines noradrenaline and serotonin , which normally act as a feedback to inhibit the release of these transmitters at the nerve endings. The concentration of the transmitters increases under the influence of NaSSA. In this respect , the effect of NaSSA is similar to that of SSNRI antidepressants ( selective serotonin and norepinephrine reuptake inhibitors ). However, they have a significantly stronger antihistaminergic and therefore sedating effect.

NaSSA are antagonists of presynaptic auto-receptors of the alpha-2 adrenoceptor type and thus cause an increased release of noradrenaline. In addition, they are antagonists of specific serotonin receptors, usually 5-HT 2A and 5-HT 2C , but also 5-HT 1F , or 5-HT 2B, depending on the NaSSA active ingredient. Mirtazapine presumably acts as an agonist on the serotonin autoreceptors of the type 5-HT 1A and thus prevents an increase in serotonergic transmission mediated by α 2A / C blockade. Usually NaSSA act as H1 antihistamines as sedating.

The first NaSSA mianserin was patented by Organon ( Tolvin ® ) in 1967 and is commercially available as a generic. Other NaSSAs are mirtazapine and the setiptiline used in Japan , both of which are chemically closely related to mianserine.
