POEM @ home ( P rotein O ptimization with E nergy M ethods, Protein Optimization with Energy Methods) was a volunteer computing the project Karlsruhe Institute of Technology , which means the technique of distributed computing , tried protein structures to optimize.

To do this, it used an atomic model for the free energy of the proteins , so POEM @ home worked for both new protein folds and applications in nanobiotechnology for which no experimental data were available. The project started in October 2007. The basis of the calculations was the BOINC software from the University of California, Berkeley . In the end, the computing power of the project was around 739 TeraFLOPS , which could fluctuate depending on the daily performance.

Scientific relevance

A different scientific approach to protein folding was used than in similar projects. For this purpose, Christian B. Anfinsen's Nobel Prize-winning thermodynamic hypothesis was used, which states that proteins have minimal free energy in their biologically active state. The otherwise usual simulation processes, which are expensive in terms of computing time, have been replaced by much faster optimization processes.

After a short time POEM @ home was able to show the first results. With these results, the project also took part in the international research competition CASP , which takes place every two years .

POEM @ home pursued the following goals:

  • Prediction of the biologically active structure of proteins
  • Understanding of the signal processing mechanisms during the interaction of different proteins
  • Understanding of disease on the protein malfunction or - aggregations based
  • Development of new drugs based on three-dimensional structures of biologically important proteins
