TBS-T buffer is based on the TBS buffer and is used to examine the binding and storage of antibodies as well as for immunodetection , including in Western Blot , in ELISA , in ELISPOT , in immunofluorescence and in immunohistochemistry . TBS-T buffer mostly consists of 10 - 50 mM TRIS as a buffer substance, 150 mM sodium chloride for a physiological ionic strength and as a surfactant 0.05% (m / V) Tween 20 , occasionally 0.1% (m / V) instead Triton X-100 . The pH is, depending on the protocol with dilute hydrochloric acid adjusted to 7.2 to 7.6. The surfactant contained reduces non-specific interactions when the antibodies or immunoconjugates are used, which results in less coloration of the background.
