The second member of the Wiskott-Aldrich syndrome proteins , also called WAVE2 , is a regulatory protein that is encoded in humans by the WASF2 gene on chromosome 1 on nine exons . It is ubiquitously expressed .
Structure and function
Like all WAVE proteins, the protein N -terminal contains a WHD (WAVE homology) domain , which is important for activation. In the direction of the C terminus there is a basic region and a proline-rich domain to which SH3 -containing signal proteins and profilin can bind. Furthermore, the VCA domain ( Verprolin homology, Coflin homology and acid = "acid"), which is present in all WASP / WAVE proteins , is crucial for the regulation of the actin cytoskeleton . This domain can namely bind to the Arp 2/3 complex and activate it; the complex in turn promotes the formation of actin filaments via nucleation . WAVE2 thus has an influence on the shape, mobility and function of the cell.
activation
Fundamental is the binding of Sra-1 (or its isoform PIR121 ), Nap-1 , Abi-1 or 2 , and HSPC300 to the WHD domain of WAVE2. It is Sra-1 a Rac effector, specifically the activated GTPase Rac recognizes and binds, which is probably the activation of WAVE2 result via the other mentioned proteins. WAVE2 thus acts as a kind of link between signal proteins and the actin cytoskeleton.
In addition, interactions of the polyproline domain of WAVE with SH3 -containing proteins or the interaction of the SH3 -domain of the Abi -proteins with proline-containing motifs of other proteins are also conceivable.
