The Kirke family is a junior branch of a family of Nottinghamshire landed gentry and descends also from the Gibson-Craig baronets.[5][6] Kirke's maternal grandmother was Israeli.[7]

Kirke was trained musically by her father Simon, the drummer for Free and Bad Company. From the age of nine, she sang in choirs, operas, and plays before moving to New York City, where she attended Fiorello H. LaGuardia High School to study classical voice and piano.

Domino became a birth doula[16] around 2009 and co-founded a doula business collective, Carriage House Birth.[17] In November 2021, she, along with entrepreneur Joanna Griffiths, released Life After Birth: Portraits of Love and the Beauty of Parenthood, a book about the experiences of postpartum mothers.[18]
