Integrin α-4 (synonym CD49d ) is a surface protein from the group of integrins .

properties

Integrin α-4 is a cell adhesion molecule . Together with integrin beta-1 or integrin beta-7, it forms a heterodimeric receptor for fibronectin and VCAM1 . It binds to the amino acid sequence GluIleAspSer (QIDS) in fibronectin. Integrin α-4 is glycosylated and phosphorylated . In contrast to other alpha integrins, integrin alpha-4 does not contain an I domain and is not activated by proteolysis . The complex of alpha-4 and beta-1 is referred to as VLA-4 and binds to fractalkine and PLA2G2A at different binding sites, whereby the binding to PLA2G2A increases the affinity of the binding of fractalkine. The complex of alpha-4 and beta-7 binds the sequence LeuAspThr in MADCAM1 . Integrin α-4 is involved in the development of chronic lymphocytic leukemia . The antibody natalizumab binds to integrin α-4 and is used to treat multiple sclerosis .

Integrin α-4 binds to LGALS8 and paxillin .
