The carboxypeptidase B2 (synonym TAFI , for T hrombin a ktivierbarer F ibrinolyse I nhibitor, carboxypeptidase U , CPU, plasma carboxypeptidase B , pCPB) is a protein in the blood, which is the blood coagulation promoted in which it the dissolution of blood clots ( fibrinolysis inhibits) .
Functions
Blood clotting
When blood clots dissolve ( fibrinolysis ), C -terminal lysine residues are formed which, as part of a positive feedback, significantly increase the activity of plasminogen and tPA . This feedback is limited by TAFI by cleaving off the C-terminal lysine residues. TAFI is activated , among other things, by thrombin (factor II). If the activity of TAFI exceeds a certain limit, it inhibits fibrinolysis. If the activity of TAFI falls below a certain limit value , the C-terminal lysine residues accumulate and thus strong fibrinolysis occurs.
Anti-inflammatory
TAFI also inhibits the activity of complement component C5 , osteopontin and bradykinin which play an important role in inflammatory reactions. TAFI has an anti-inflammatory effect.
