Structure of the ctRNA p42d

ctRNA ( English counter-transcribed RNA , counter-transcribed RNA ') are RNA from bacteria , with which the number of plasmids in a bacterial cell is limited. The inhibition takes place when the plasmid replicates . Limiting replication by ctRNA is a mechanism for limiting the number of plasmid copies, e.g. B. to avoid excessive gene expression of the genes on the plasmid.

properties

ctRNA are non-coding ribonucleic acids , attached to the mRNA of repB bind and its Translation inhibit. RepB is a protein that initiates the rolling circle replication of plasmids. By inhibiting the transcription of RepB , the number of plasmid copies in the bacterium is reduced. The ctRNA-containing plasmids are therefore referred to as low copy number plasmids .

In Corynebacterium glutamicum , ctRNA serves as an antisense RNA . In Enterococcus faecium , the plasmid pJB01 contains the three open reading frames copA , repB and repC and on the opposite strand between copA and repB is the sequence of the pJB01-ctRNA, which binds to the three open reading frames and inhibits their translation. Further ctRNAs have been described in lactococci, Streptococcus thermophilus , and Rhizobium etli .

ctRNA, iterons and antisense RNA are bacterial mechanisms that limit the number of plasmid copies.
