Response type Cleavage of double-stranded DNA
Substrate X-GGCC-Y (DNA) + H 2 O
Products X-GG + CC-Y
Occurrence
Parent taxon Haemophilus aegyptius

The enzyme HaeIII belongs to the restriction endonucleases and is able to cut DNA at specific points. HaeIII is a type II restriction enzyme and is the third restriction enzyme isolated from the bacterium Haemophilus aegyptius . The coding gene has been sequenced and cloned, which means it can be used for many applications in molecular biology. The molar mass of the enzyme is 37,126  Da .

Like many other restriction enzymes, HaeIII binds to sites in DNA that have a specific nucleotide sequence.

Recognition sequence Restriction cut
5'-GGCC-3'
3'-CCGG-5'
5'-GG	CC-3'
3'-CC	GG-5'

When cutting the double-stranded DNA, a smooth cut without overhangs is created. The ends of the DNA fragments are therefore referred to as blunt ends .
