The Missouri State Route runs eight miles through the northeast of the state. The southern terminus is on US Highway 61 5.8 km south of Wayland . After about one kilometer, US Highway 61 leaves the joint route to the east and Missouri State Route 27 runs alone to the north.
With the place Wayland the street passes the only larger town and crosses the US Highway 136 there . A few kilometers to the north, Missouri State Route 27 ends on the border between the states of Missouri and Iowa on the Des Moines River .
Beyond the bridge over the river, an identically numbered road, Iowa Highway 27 , continues north.
Missouri State Route 27 and Iowa Highway 27 are part of the Avenue of the Saints, along with a number of other numbered highways.
