The regulator protein Rop is a small protein , the gene naturally in bacteria - plasmids occurs and the replication regulates the plasmid in the respective bacteria. For this purpose it binds to the RNA I which modulates the primer (RNA II), which in turn initiates the replication and thereby changes the initiation rate of the replication. Rop or its gene is used biotechnologically in the artificial plasmid pBR322 .

The coding region for the so-called Rop ( repressor of primer ) or ROM protein ( RNA I Modulating protein) is outside the actual origin of replication of pBR322 - plasmid . This is a 7.2 kDa major protein consisting of 63 amino acids which comprises, hybridization between RNA catalyzes I and RNA II. By knockout of ROP, the copy number of ColEI plasmids can be increased.
