TlTx1 (synonym Kappa-theraphotoxin-Tb1a ) is a protein and neurotoxin that is produced by the giant tarantula Theraphosa blondi ( synonym : T. leblondi ).

Disulfide bridges of TlTx1

TlTx1 is formed in the venom glands, like TlTx2 and TlTx3 . It inhibits voltage-gated potassium channels of the type Kv4.2 / KCND2 with an IC 50 of 193 nM by shifting the activation of the channel towards stronger depolarization . Presumably, TITx1 binds to the S3-S4 linker in the voltage-dependent protein domain of the channel. It is related to heteropodatoxins and phrixotoxins . It has three disulfide bridges .

The toxins TlTx1-3 can be distinguished by mass spectrometry and by Edman degradation .
