The transcription factor IIB (TFIIB) is a protein which in the cell nucleus of vertebrates in DNA binding and the initiation of transcription participates. In particular, TFIIB is responsible for recognizing any BRE element that may be present in the core promoter of the respective gene . During the assembly of the pre-initiation complex , TFIIB binds to the promoter-TFIID-TFIIA complex (DA complex) and thus forms the DAB complex, to which RNA polymerase II docks. TFIIB contains a zinc finger domain for DNA binding .

TFIIB interacts with the center of activity of RNA polymerase. This may prevent the polymerase from docking with the DNA without the pre-initiation complex, thus ensuring positioning accuracy.

TFIIB is also able to form complexes with TCEA2 , GPBP1 , the HIV protein VPR and the Epstein-Barr virus protein EBNA2.
