The WW , WWP domain or also Rsp5 domain is a protein domain of different proteins . It interacts with various protein ligands . It can also bind to proline- rich domains / proteins, to [AP] -PP- [AP] -Y and to domains that contain phosphoserine - phosphothreonine .

Structure and occurrence

The WW domain folds into a stable, three-stranded β-sheet . It is relatively short with just 38 amino acids and is repeated up to four times in certain proteins such as in human NEDD4 . Its name is derived from the presence of two tryptophan residues (abbreviated with "W"), which are located about 20-23 amino acids apart and which occur in most of the known WW domains. Hydrophobic residues form a flat spot that serves to bind with proline.

The WW domain was first discovered when the two connected isoforms of YAP (“Yes associated protein”) YAP1-1 and YAP1-2 were examined more closely. YAP1-2 differs from YAP1-1 in that it has 38 more amino acids. These 38 amino acids were called the WW domain (actually they represent the second WW domain of YAP1-2 ).
