Guido I of Lusignan (* around 1265; † September 24 or November 28, 1308 in Angoulême ) was Lord of Lusignan , Count of La Marche and Angoulême .

He was the son of Hugo XII. by Lusignan and Johanna von Fougères. After his father's death in 1270, he succeeded him as Lord of Couhé and Peyrat. His older brother Hugo XIII. von Lusignan inherited the paternal rule of Lusignan and the counties of La Marche and Angoulême. When Hugo XIII. 1303 died childless, Guido was his successor.

Guido died unmarried and childless in 1308. His sister Jolanda succeeded him as mistress of Lusignan, Couhé and Peyrat and Countess of La Marche. The county of Angoulême was sold by Guido's sisters Johanna and Isabella to the French king Philip IV , who added it to the crown domain .
