Macrophomat synthase is one of the few natural enzymes believed to be able to catalyze a pericyclic reaction . In the case of Macrophomat synthase, it is more precisely a Diels-Alder reaction .

So far, the enzyme is only known from the fungus Macrophoma commelinae and is also named after it. It consists of 339 amino acids and is only active in the presence of magnesium (2+).

The product of the Diels-Alder reaction of oxaloacetate and a substituted 2-pyrone catalyzed by this enzyme is what is known as the macrophomat after carbon dioxide has been split off twice . It is a multiply substituted benzoic acid derivative.

In the absence of the 2-pyron, macrophate synthase functions as a decarboxylase with high catalytic activity.
