The place is located on the second largest and most populous island of Santa Cruz in the Galápagos Archipelago. Since UNESCO drafted a nature conservation plan for the islands in 1957, the population of Puerto Ayora has increased five hundredfold, from 30 to around 12,000 people in 50 years. The Charles Darwin Research Station is located here .

Puerto Ayora is the seat of the Municipalidad de Santa Cruz , which as Dirección Cantonal (regional authority ) also manages the other settlements on the island of Santa Cruz and the non- military parts of the neighboring island of Baltra . The modern administration building was built on Calle Seymour.

Most of the hotels, bars and restaurants are on Avenida Charles Darwin by the sea, which is also where the hospital, town hall ( Alcaldía ) and the modern Iglesia de San Francisco church are located. It was built in 1968. The main shopping streets with numerous retail and specialty shops are Calle Baltra and Calle Durán, a little inland . In the northern part of the city, in addition to various schools, a market hall, a sports center, a stadium, a health center ( Centro de Salud ) for outpatient treatment and two other modern churches were built.
