The Strait of Semau is a strait in the Timor Archipelago of the Lesser Sunda Islands . It separates the island of Semau in the west from the island of Timor in the east. At the same time it connects the Sawu Sea in the north with the Strait of Roti in the south. The small island of Kambang lies in the strait off the coast of Semau . At the north end of the road on the Timor side is the entrance to Kupang Bay .

Politically, the region belongs to the Indonesian province of East Nusa Tenggara .
