His brother-in-law Francis Tresham was a co-conspirator of the Gunpowder Plot . On October 26, 1605, William Parker received a letter advising him to stay away from the opening of Parliament on the pretext that the house would "receive a blow". He passed this letter on to Robert Cecil, 1st Earl of Salisbury , who reported it to the king. On the morning of November 5, 1605, he took part in the inspection of the cellars under Parliament, where the explosives stored there were discovered. The king gave him money and land as a reward for his loyalty.

At his death in 1622 his son Henry inherited his titles.
