Kinyoun staining

Mycobacterial infection of the small intestine , stained according to Kinyoun.

The Kinyoun stain is a histological stain of microorganisms with an acid-fast cell wall , especially mycobacteria , nocardia and some protozoa .

It is named after the first director of the US American hygiene laboratory on Staten Island, Joseph J. Kinyoun (1860-1919).

properties

The Kinyoun stain is similar to the Ziehl-Neelsen stain , but does not require the slides to be heated . The red color is achieved with carbol fuchsin , followed by decolorization in an acidic alcoholic solution and a second coloration with methylene blue . The use of carbol fuchsin in place of basic fuchsin does not require heating. The red color indicates organisms with an acid-fast cell wall, while all other cells are counterstained with methylene blue. As an alternative to one percent sulfuric acid, a three percent hydrochloric acid solution can be used. With sulfuric acid the discoloration is slower, which is why it is used to identify weakly acid-fast microorganisms such as nocardia. As an alternative to methylene blue, brilliant green can be used as a second color. The Kinyoun staining can be combined with the Gram staining ( Gram-Chromotrop-Kinyoun-staining , GCK).

As an alternative to the Kinyoun staining, an auramine - phenol staining can sometimes be carried out.
