The Newfoundland and Labrador Route 100 is a highway on Newfoundland in the east of the island. It opens up the area south of the Trans-Canada Highway near Whitbourne . The northern section is part of the National Highway System and is designated as a core route.

The route begins on Route 1 west of Whitbourne. It leads to the southwest and is called the Argentia Access Road on this section of the route. This route leads to Argentia, a part of Placentia ; in Argentia there is a commercial port next to an important industrial area. Until 1994 there was also a military base with a port and airport. From Placentia the route runs as a coastal road west along the southern foothills of the Avalon Peninsula to St. Bride's , where the road changes to the east coast to Branch . There it meets Route 92 , which runs along the east coast when coming from the north.
