The Newfoundland and Labrador Route 40 is located on the Avalon Peninsula in the Canadian province of Newfoundland and Labrador . It has a length of approx. 15 km and connects the provincial capital St. John’s with the coastal community Portugal Cove-St. Philip’s . Due to the connection to the international airport of the city of St. John's, the route belongs to the National Highway System , the section between the airport and the Trans-Canada Highway is classified as a main route (core route).

The route begins in a residential area in St. John's and heads north. It crosses the Trans-Canada Highway and serves as a connection for St. John's International Airport. From there the route runs west to the coast and ends in Portugal Cove at Conception Bay . There is a ferry terminal that connects Bell Island with Newfoundland.
