After the death of his father in 1644 he inherited parts of Ledóchow and Krasnopol, from 1667 he was the owner of Koryt, Chocim and Ledkow. In 1638 he enrolled with his brother Marcin Ledóchowski as a student at the University of Cracow . He then served in the army and was stationed in Ukraine . In 1648 he was captured by the Tatars at the Battle of Korsun and remained in it for two years.

In 1658 he received the post of judge (Podkomorzy) of Kremenec. In 1661 he entered the Reichstag as envoy of the Kremenec district and as commissioner oversaw military spending. In the Reichstag he strongly opposed the election of a king vivente rege (during the lifetime of an incumbent king) in order to prevent the emergence of an absolutist hereditary monarchy. In 1661 he was commissioned by the Reichstag to sign contracts with Moscow as commissioner. In this capacity he took part in negotiations in Lviv in 1663. On April 26, 1664, he and Cyprian Brzostowski signed a provisional agreement between Moscow and the Republic of Poland-Lithuania. He conducted further negotiations in Andruszow in 1666 and 1667. 1670 led to a diplomatic mission to Moscow. In 1673 he became castellan of Braclav and a member of the royal council of war.

In 1674 he signed the election of Jan Sobieski as King of the Republic, whereupon the latter entrusted him with the office of castellan of Volhynia shortly after his coronation. In the same year, the Queen's widow Eleonora of Austria asked Stefan Ledóchowski for support in her efforts to travel to Silesia. Until his death in 1676, Stefan Ledóchowski was one of the king's trusted advisers on questions relating to Ukraine.

In the course of his life he managed to amass a considerable fortune. He acquired the crown lords of Plaszow and Ceceniowec and received part of the possessions of King Michał Korybut Wiśniowiecki . He was also the patron of the Dominican monastery in Podkamien.

In 1656 he married Anna Srzedzinska and had two daughters and four sons with her, including Felix Ledóchowski and Stanislaw Ledóchowski , Marshal of the Tarnogrod Confederation, who both took part in the liberation of Vienna from the Turks in 1683.
