He was sheriff of Glamorgan from 1593 to 1594, from 1603 to 1604 and from 1622 to 1623 . In addition to numerous other offices, he succeeded his father from 1585 until his death, Chamberlain and Chancellor of South Wales. In the 1597 general election, he was elected Knight of the Shire for Glamorgan. After first John Herbert in 1601 and then Philip Herbert were elected Knight of the Shire in 1604 , Mansel was again Knight of the Shire by by-election in 1605, after Philip Herbert had been elevated to Earl of Montgomery . In the general election in 1614 Mansel was re-elected. Several times, so in 1606 and 1610, he was a member of delegations from the House of Commons , who complained to King James I about his government. Thomas Mansel was one of the 200 members of the gentry who were first raised to the rank of baronet on May 22, 1611 by Letters patent , he received the title of Baronet, of Margam in the County of Glamorgan .

After his wife died before 1603, he married Jane Bussy , the widow of John Fuller and John Bussey , in 1603 in second marriage . Through this marriage he acquired considerable real estate in Lincolnshire . He had a daughter with his second wife:

He was buried in Margam Abbey. His heir became his eldest son, Lewis Mansel . His third eldest son Arthur Mansel married Jane Price , the daughter and heiress of William Price, and thus acquired Briton Ferry at Neath . Arthur's son Bussy probably got his unusual first name after Jane Bussy, the second wife of his grandfather Thomas.
