The colipase is approximately 12 k Da large protein , which as a cofactor for the function of pancreatic lipase is necessary. It is formed as an inactive precursor ( procolipase ) in the pancreas , which is cleaved in the small intestine by trypsin into colipase and the N-terminal pentapeptide enterostatin .

The colipase forms the prolipase-pancreatic lipase complex with the pancreatic lipase and mediates the contact of the lipase with the lipid interfaces consisting of phospholipids or bile acids . In addition, the colipase causes a conformational change in the lipase, which opens the lid over the active center of the lipase.
