In 1848, the then Wardapet Makar Ter-Petrosian transferred a considerable part of the manuscripts of the monastery of St. Stephanos ("Maghardavank"; Iranian East Azerbaijan), including the Etschmiadzin Gospels , to the library of the Echmiadzin monastery; From 1849 to 1851 he acted as its director.

In 1852 Makar was ordained bishop by Catholicos Nerses V and in 1854 he was made archbishop. From 1864 to 1874 he worked as bishop of the Armenian diocese of Georgia and Imereti (based in Tbilisi ), from 1874 to 1885 of the diocese of Nor-Nakhichevan in southern Russia. Since the tsarist authorities rejected the election of a candidate of Ottoman nationality, Makar I was anointed Catholicos on November 10, 1885, after several years of vacancy. Contrary to Russian expectations, he held office for only a few years, resolutely defending Armenian interests, died in 1891 of the consequences of a heart attack and was buried in front of the Surb Etschmiadzin cathedral.
