William Shuford Sly (born October 19, 1932 in East St. Louis ) is an American biochemist and molecular biologist .

Sly earned an MD from Saint Louis University in 1957 . Since 1984 he has been Professor of Biochemistry and Molecular Biology there .

Sly and co-workers made important contributions to the elucidation of the biochemical mechanisms by which enzymes reach their respective destination from the endoplasmic reticulum within the cell . Lysosomal enzymes carry mannose-6-phosphate residues, which act as transport signals. Further work deals with the pathogenesis of lysosomal storage diseases .

Mucopolysaccharidosis VII was named Sly syndrome after William S. Sly . Sly's work led to enzyme replacement therapy for Gaucher's disease .

Sly became a member of the National Academy of Sciences in 1989 . In 1991 he and Stuart Kornfeld received the Passano Award .
