Dorothea was the eldest child of Elector Friedrich II of Brandenburg (1413–1471) from his marriage to Katharina (1421–1476), daughter of Elector Friedrich I of Saxony .

She married Duke Johann IV of Saxony-Lauenburg (1439–1507) on February 12, 1464 in Lüneburg . As the eldest daughter of Elector Friedrich, who had no surviving sons, the marriage speech is important. In addition to 10,000 gulden bridal treasure , Friedrich promised his son-in-law: “ So everything that our dear daughter of erbs and right because of all our hereditary possessions, whether or not we can inherit from us, may be born according to antzal as vil as the other unnser children, one who lives in Well Inn seyen, and which the almechtig goth might still forbid us ”. However, Friedrich later resigned in favor of his brother Albrecht Achilles in order to keep the inheritance in the family, according to his own admission.

Friedrich also owed his son-in-law the marriage money. Dorothea's uncle Johann therefore compared himself to Duke Johann in 1482.

On March 20, 1519, Duchess Dorothea found her final resting place in the Ratzeburg Cathedral . The preserved tombstone shows her and her husband in relief in round arch niches.
