He was the owner of Norddeutsche Handelsgesellschaft, Mackenthun & Co. in Lamspringe , a supplier for the electrical engineering industry in Germany and abroad. In addition, he was the owner and director of Niedersachsenwerke AG, which manufactured and sold porcelain and electrical goods. In 1925 the two companies merged to form Hannoversche Porzellanfabrik & Metallwerk AG, which had to cease operations in 1934. In addition, Mackenthun was the owner of a Lamspringen electricity company and farmer. From 1927 he lived in Holzminden .

After the First World War Mackenthun joined the German Democratic Party and became district chairman of the party in the Hildesheim administrative district . In addition, he was a senator and a member of the municipality of Lamspringe. In February 1921 he was elected as a member of the Prussian state parliament, where he represented constituency 15 East Hanover. During his parliamentary work, he only took part in seven of 124 votes, missed 46 without excuse and was reported sick in a further 71. Due to his breach of duty, the DDP parliamentary group urged him, at the instigation of the second parliamentary group chairman Walther Schreiber, to give up his mandate, which he resigned on January 22, 1924 after his previous refusal. For him, drew Mathilde Drees by the parliament.
