James Hamilton, 2nd Marquess of Hamilton KG (* 1589 - March 2, 1625 in Whitehall ) was a Scottish nobleman .

James Hamilton was a son of John Hamilton, 1st Marquess of Hamilton and his wife Margaret Lyon. After the death of his father, he followed in 1604 as 2nd Marquess of Hamilton . In 1609 he became the heir to his uncle, James Hamilton , 4th Earl of Arran . In 1619 he was also raised to 1st Earl of Cambridge . He held the office of Lord High Commissioner to the Parliament in Scotland from 1621 and from 1624 to 1625 he was Lord Steward of the Household.

At times there was speculation about a marriage between James Hamilton and Princess Elisabeth . Rumor has it that the marquess's death was also due to poisoning.

James married Anne, daughter of James Cunningham, 7th Earl of Glencairn, and had the following children with her:
