Walther was born the daughter of an officer . She discovered her comedic talent and her passion for the theater early on and, after attending Christianshavns Gymnasium, studied from 1953 to 1956 at the Privatteatrenes elevskole , the drama school of the New Theater in Copenhagen. After a first film appearance in 1956, she became known in 1957 for her role as Tjenestepige in Annelise Reenberg's film Tre piger fra Jylland . Although she appeared in many movies and two television series ( Huset på Christianshavn (“ Oh, these tenants !”), 1970–1977, and Smuglerne , 1970) in the following three decades , she stayed with the theater - and especially with the Copenhagen ABC Teatret  - always loyal.

Kirsten Walther became popular and particularly well-known in Germany , alongside Ove Sprogøe , Poul Bundgaard and Morten Grunwald in the Danish film series Die Olsenbande , which was directed by Erik Balling . Here she played the role of Yvonne, the naive and constantly nagging wife of the small crook Kjeld, who became more and more of a female lead over the course of the films. Three years after her last film appearance in The Olsen Gang Flies Over All Mountains (1981), she withdrew from acting in order to devote herself to her family. On February 19, 1987, Kirsten Walther died unexpectedly of heart failure at the age of 53. She found her final resting place in the "Vedbæk Kirkegård" cemetery in Vedbæk on the northern edge of Copenhagen . Next to her grave, her fellow actor Morten Grunwald, who died in 2018, was buried. After Walther's death it seemed impossible to continue the Olsen Gang series. It wasn't until 1998 that the last part of this popular series was filmed without them.

Kirsten Walther was married to the director and producer Palle Ruben Wolfsberg (1931–2019) since September 26, 1959 . She and her husband adopted a two-year-old boy from Greenland in 1966.
