L- arginine: glycine amidinotransferase (AGAT, EC ) is an enzyme that is involved in the synthesis of guanidinoacetate from the amino acids arginine and glycine , mainly in the kidneys and salivary glands.

The L- arginine: glycine amidinotransferase catalyzes the following reaction:
L- arginine + glycine ↔ L- ornithine + guanidinoacetate
Guanidinoacetate is the direct precursor of creatine in the metabolism . The L- arginine: glycine amidinotransferase is produced in the brain , in the heart , in the kidneys (highest gene expression ), in the liver , in the lungs , in the salivary gland , in the placenta , in a fetus and in the skeletal muscles . The Michaelis-Menten constant is 2.0 µM for arginine, 3.0 µM for glycine and the maximum reaction rate is 0.44 µmol / min / mg enzyme.
