Pläschke, fourth of seven children of the builder and shopkeeper Samuel Gottlieb Plaeschke (* 1786) and his first wife Johanna Rosine, née Baumann (1812-1846), attended the Düsseldorf Art Academy from 1836 to 1840 after an apprenticeship in Breslau . There Theodor Hildebrandt became his most important teacher. As a freelance genre painter, he settled permanently in Düsseldorf in 1844. The painter Moritz Blanckarts was his private student in his studio . In 1846 Pläschke married Mathilde Knevels († in November 1857), who gave birth to the children Johann Gottlieb Moritz (1847–1914), Alfred (1849–1907) and Eugen (1851–1898). In the revolutionary year of 1848 he was one of the founding members of the artists' association Malkasten , to which he belonged until his death.

On March 31, 2019, an episode of the NDR's Lieb & Teuer program was broadcast, moderated by Janin Ullmann and filmed in Reinbek Castle . In it, an oil painting by Pläschke was discussed with the painting expert Ariane Skora, which shows a pair of siblings and can be ascribed to genre painting.
