Protein P is a protein from the group of reverse transcriptases and is produced by the hepatitis B virus .

properties

Protein P is bound to viral RNA in the cytosol within the capsid of HBV from HBcAg . The RNA is then transcribed into DNA by protein P. As a reverse transcriptase, Protein P is an RNA- or DNA-dependent DNA polymerase . Protein P also has a 3 ': 5' endonuclease activity . For reverse transcription, protein P binds to the epsilon loop of pregenomic RNA, protein P itself serves as a primer and is therefore covalently linked to the 5 'end of the (-) DNA. The (+) DNA is then generated using the (-) DNA template. After budding and a renewed infection of a cell, the double-stranded DNA with the capsid is imported into the cell nucleus , where the double-stranded DNA is closed to form cccDNA and the protein P is separated from the genome.

Applications

Lamivudine , penciclovir , tenofovir , adefovir and entecavir inhibit protein P of non-resistant HBV. Protein P is also being investigated for the treatment of HBV infections.
