The Rivière Mistassini is a small river in the Côte-Nord administrative region of the Canadian province of Québec .

The Rivière Mistassini has its origin 10 km east of the Réservoir Manic 2 . It flows through the regional county town of Manicouagan in the coastal region in the extreme south of the Labrador Peninsula in a predominantly southeastern direction. It flows through Lac Mistassini and other smaller lakes. Finally, it flows into the estuary of the Saint Lawrence River at Pointe Mistassini 4 km west of the Franquelin settlement . Just before the estuary, Route 138 crosses the river. The Rivière Mistassini has a length of 33 km.
