Sucrose-epichlorohydrin copolymer is a synthetically produced copolymer from sucrose and epichlorohydrin that dissolves easily in water. The molecule is highly branched and reacts neutrally in aqueous solution .

The copolymer is used in density gradient centrifugation in biochemical analysis.

Manufacturing

Ficoll is prepared from sucrose and epichlorohydrin copolymerized . The density of the copolymer can be influenced. A density of 1 to 1.2 g / ml is used for gradient centrifugation.

use

Sucrose-epichlorohydrin copolymer is used in biological and medical research and diagnostics for the isolation of mononuclear cells ( PBMC ) under the brand names Ficoll , Histopaque or Polysucrose . As a component of the sample buffer for agarose gels , it increases the density of the DNA -containing solution brought into the sample wells, so that it stays longer in the wells. Sucrose-epichlorohydrin copolymers have practically no osmotic activity due to their high molar mass ( Ficoll 400 : on average 400  kDa ) and are therefore more suitable than sucrose, in particular for density gradient centrifugation.
